Biocolor Fastin elastin Assay
利用染料结合的方法来定量检测释放到组织培养液或从生物材料中提取的弹性蛋白。
应用范围:样本可来源于细胞培养基和含细胞外基质成分的组织提取物(如弹性软骨、主动脉、皮肤等),可检测可溶性弹性蛋白原和不可溶性弹性蛋白(抽提后以可溶性弹性蛋白多肽的形式检测)。
样本要求:样本体积5-100ul,包含5-75ug的弹性蛋白
实验操作步骤
弹性蛋白的分离:用弹性蛋白沉淀剂处理样本,使弹性蛋白沉淀下来。
弹性蛋白与染料结合:装有沉淀的弹性蛋白管中,加染料和反应液,反应60min以上
弹性蛋白-染料复合物获得:去掉未结合的染料
复合物染料的释放:添加染料释放剂,使弹性蛋白-染料复合物中的染料释放出来
释放出的染料检测:绘制标准曲线,513nm或405nm处定量检测
